Enables no-wash/no-lyse application
Immunophenotyping of whole blood using traditional flow cytometry is typically performed with lysis of the red blood cells (RBCs) followed by a wash step. These steps can result in significant cell loss and damage.
While a no-lyse, no-wash method for immunophenotyping of whole blood has been previously described, the method is of limited utility in traditional flow cytometry, which use hydrodynamic forces to orient cells in the flow stream. Moreover, significant sample dilution is required in no-lyse, no-wash methods to achieve low coincidence with red blood cells, necessitating extremely long acquisition times.
With the Attune™ NxT cytometer you can acquire data quickly, even from dilute samples, using either a fluorescent reagent as a trigger to eliminate red blood cells or a differential side scatter using both blue and violet scatter. This kit enables the side scatter approach.
